Each Transfection-Ready DNA sample undergoes quality control and comes with a Certificate of Analysis.
Concentration and Volume
All Transfection-Ready DNA is provided as liquid DNA in Tris buffer at a concentration of 1.0 µg/µL ± 2.5%. 50 µg samples are provided in a volume of 50 µL. 100 µg samples are provided in a volume of 100 µL
Whole-Plasmid Sequencing
Each Transfection-Ready DNA prep is fully sequence-verified by NGS. QC sequencing of Transfection-Ready DNA samples is performed through our collaboration with Plasmidsaurus, a pioneer of whole plasmid sequencing. Plasmidsaurus utilizes Oxford Nanopore Technology (ONT) long-read sequencing and an automated custom analysis pipeline, resulting in a basecall accuracy of 99.9999%.
Addgene scientists analyze the NGS results to validate that the sequence of the plasmid is as expected, and that there are no contaminants detected.
Endotoxin Levels
Addgene has validated its production process to ensure consistently low endotoxin levels in every Transfection-Ready DNA prep (Figure 1). Please note that individual lots are not tested for endotoxin.
Figure 1: Endotoxin levels in standard miniprep DNA vs. Addgene Transfection-Ready DNA. Transfection-Ready DNA consistently measures below 1.0 EU/µg, well under the levels seen in standard minipreps. Values are based on endotoxin testing from internal validation of Addgene’s Transfection-Ready DNA production process.
