Store the glycerol stocks in the kit at -80°C upon receipt. Take precautions to minimize freeze-thaw cycles. Keep the plate on dry ice to prevent thawing. To access a plasmid, use a sterile pipette tip to puncture the seal above an individual well and then streak a portion of the glycerol stock onto an agar plate to isolate single colonies. To patch the hole, use sterile tape or a portion of a fresh aluminum seal.
